Position Overview

The client is seeking a highly skilled and motivated Biologist to join our diabetes team. This role will play a key laboratory-based function in advancing iPSC stem cell engineering and contribute to the development of next-generation pancreatic islet cell therapies. The candidate will work under the Director of Islet Cell Therapy, Stem Cell Biology, and Cell Engineering and will be responsible for both research and translational activities in stem cell biology.

This position is located in Boston, MA, with mandatory weekend cell culture responsibilities on a rotational basis.

Key Responsibilities

  • Advance iPSC gene editing and cell line development, including evaluation of differentiated cells.

  • Perform routine iPSC cell culture, passaging, and media preparation.

  • Optimize workflows for CRISPR-based gene editing, gRNA screening, and delivery.

  • Design and perform assays for on-/off-target analysis using PCR and NGS workflows.

  • Generate and characterize clonal gene-edited iPSC lines using flow cytometry, ddPCR, dPCR, and qPCR.

  • Execute directed differentiation of human iPSCs to pancreatic islets and assess drug substance parameters such as cell counts, viability, and metabolite profiles.

  • Evaluate genomic stability of iPSC lines and drug substances, assisting team members with mitigation strategies.

  • Collaborate cross-functionally to support the transfer of R&D assays to GMP standards.

  • Maintain accurate experimental records, synthesize results, prepare reports, and contribute to SOP development.

  • Ensure timely execution of work plans and adherence to project deadlines.

Required Qualifications

  • BS or MS in Stem Cell Biology, Cell Biology, Bioengineering, or a related field.

    • Minimum 3 years of relevant experience for MS

    • Minimum 5 years of relevant experience for BS

  • Extensive hands-on experience with human iPSC culture, gene editing, and molecular biology techniques.

  • Proficiency in gene editing and cell line development, including protocol optimization and flow-based cell sorting.

  • Experience with cell delivery methods for gene editing tools.

  • Strong communication and interpersonal skills, with the ability to work effectively in a collaborative team environment.

  • Excellent organizational skills and meticulous record keeping.

Preferred Qualifications

  • Experience in pancreatic islet-directed differentiation.

  • Hands-on experience transitioning research-grade processes to GMP-grade workflows.
